The obsession is total and stated flatly: 'por maneira nenhuma,' in no way whatever, could he give her up. The verb 'desfazer-se de' — to rid oneself of, to dispose of — treats Isaura as property he refuses to relinquish, which is exactly the legal truth. His refusal to free her is at once possessive lust and the plain exercise of ownership.
